これら 2 つのホルモンのバランスは、体内の全体的なエネルギーバランスを達成するために必要です。肥満では、レプチンに対する感受性が低下します(アンダーバーグら、2016)。これは大きな問題であり、体の他の部分に多くのエネルギーが蓄えられているにもかかわらず、脳が満腹感を検出できなくなることになります。なぜこのようなことが起こるのでしょうか? 肥満のヒト被験者におけるレプチン耐性の根拠は不明です。過食によりレプチンレベルが持続的に上昇したままの場合、レプチン受容体のダウンレギュレーションが発生し、その結果ホルモンに対する感受性が低下する可能性があります。人間において、そして実際に他の動物においても、低カロリー食によってレプチンレベルが低下すると、血漿レプチン濃度が低下し、高レベルの継続的な空腹感が引き起こされます。これがダイエットの失敗率の高さを説明しているのかもしれません。レプチンレベルが低いと、体重増加に対する強力な刺激となる可能性があります。
